Pharmacological action
Antithyroid drug. It blocks the enzyme peroxidase involved in the iodination of thyroid hormones of the thyroid gland, which leads to a violation of the synthesis of thyroxine and triiodothyronine.
Pharmacokinetics
After oral administration, it is well absorbed from the digestive tract. T1 / 2 is 6 hours. Biotransformed in the body.
Excreted in the urine as metabolites (70% for 48 hours) and unchanged substance.
Indications
Diffuse toxic goiter
thyrotoxic crisis
mixed toxic goiter (as part of combination therapy with thyroid hormonal drugs)
as part of combination therapy with radioactive iodine.
Contraindications
Severe leukopenia or granulocytopenia
hypersensitivity to thiamazole
pregnancy
lactation (breastfeeding).
Use during pregnancy and lactation
Contraindicated in pregnancy and lactation (breastfeeding).
Thiamazole crosses the placental barrier. Application after the first trimester of pregnancy is associated with the threat of hypothyroidism in the fetus.

Dosage and administration
Installed individually. The usual dose is 20-40 mg / day in 2-4 doses, depending on the severity of the disease. After normalization of thyroid function (usually after 3-8 weeks), maintenance doses of 2.5-10 mg / day are used.
For children - 300-500 mcg / kg, maintenance doses - 200-300 mcg / kg. In maintenance doses, thiamazole can be taken 1 time / day (in the morning after a meal).
Side effects
Allergic reactions: skin rash rarely - fever in isolated cases - lupus-like syndrome.
From the hematopoietic system: rarely - hematopoiesis disorders (leukopenia, agranulocytosis) in isolated cases - generalized lymphadenopathy.
From the digestive system: rarely - taste impairment in isolated cases - impaired liver function, nausea, vomiting,
From the central nervous system and peripheral nervous system: in isolated cases - neuritis and polyneuritis, headaches.
From the endocrine system: thyroid hyperplasia.
Drug Interaction
Concomitant use of thiamazole with amidopyrine, sulfonamides increases the risk of leukopenia.
Leukogen and folic acid, when co-administered with thiamazole, reduce the risk of leukopenia.
Gentamicin enhances the anti-thyroid action of thiamazole.
Storage Conditions
