Pharmacological action
Wazonite - angioprotective, antiplatelet, vasodilator.
Improves microcirculation and rheological properties of blood. It inhibits phosphodiesterase, contributing to the accumulation of cAMP in tissues and organs, including in the cells of the smooth muscles of blood vessels, in the formed elements of the blood.
It inhibits platelet and erythrocyte aggregation, increases the elasticity of their membranes, lowers plasma fibrinogen concentration and enhances fibrinolysis. Improves the supply of tissue with oxygen, most in the limbs, central nervous system and, to a lesser extent, in the kidneys. Slightly dilates the coronary vessels.
Pharmacokinetics
When ingestible tablets are administered, the active substance is continuously released and absorbed uniformly from the digestive tract. It undergoes metabolism in the liver during the "first passage", as a result of which a number of pharmacologically active metabolites are formed.
Cmax of pentoxifylline and its active metabolites in plasma is reached after 3-4 hours and remains at the therapeutic level for 12 hours. It is excreted mainly with urine in the form of metabolites.

Contraindications
Hypersensitivity to pentoxifylline and other xanthine derivatives
acute myocardial infarction

Dosage and administration
Enteric-coated tablets are swallowed whole with a little water.
The daily dose is divided into 3 doses. The initial dose is 600 mg / day.
As the condition improves, the dose can be reduced to 300 mg / day. Prolonged dosage forms are prescribed 2-3 times a day.
Side effects of the
From the digestive system: possibly nausea, vomiting, diarrhea, dry mouth, decreased appetite, sensation of fullness in the abdomen, epigastric pain, increased activity of liver enzymes (ALT, AST, LDH) and alkaline phosphatase, cholecystitis, hepatitis, jaundice rarely - an unpleasant taste in the mouth, hypersalivation.
From the side of the central nervous system: relatively rarely - headache, dizziness, anxiety, sleep disturbances, cramps.
On the part of the sensory organs: rarely - conjunctivitis, scotoma, blurred visual perception, earache.
From the cardiovascular system: when using the drug in high doses, a decrease in blood pressure, tachycardia, angina pectoris, heart rhythm disturbances, hyperemia of the skin of the face, flushing of the skin of the face and upper chest, swelling.
Allergic reactions: possibly - itching, skin rash, urticaria, Quincke's edema rarely - anaphylactic shock.
From the hemopoietic system and hemostasis: rarely - bleeding (from the vessels of the stomach, intestines, skin and mucous membranes), thrombocytopenia, leukopenia, pancytopenia, hypofibrinogenemia, changes in the pattern of peripheral blood, aplastic anemia. Pentoxifylline may enhance the action of drugs that affect the blood coagulation system (indirect and direct anticoagulants, thrombolytics), anitibiotics (including cephalosporins - cefamandol, cefapetazole, cefapetazole, cefatrazole).
Increases the effectiveness of hypotensive drugs, insulin and oral hypoglycemic drugs.
Cimetidine increases the concentration of pentoxifylline in plasma (risk of side effects).
Joint appointment with dr. xanthines can lead to excessive nervous excitation of patients.
Overdose
Symptoms: nausea, dizziness, decrease in blood pressure, fever, tachycardia, drowsiness or agitation, loss of consciousness, areflexia, tonic-clonic convulsions, vomiting, like arrhythmias.
Treatment: gastric lavage, then carry out symptomatic therapy. There is no specific antidote. In case of vomiting with traces of blood, gastric lavage is not allowed.
