pharmachologic effect
Pancreatic hormone. The mechanism of action of glucagon is due to its binding to specific receptors of liver cells, which leads to an increase in G-protein-mediated adenylate cyclase activity and an increase in the formation of cAMP. This enhances the catabolism of deposited glycogen and activates gluconeogenesis and ketogenesis. Glucagon has no effect on muscle glycogen, apparently due to the lack of glucagon receptors. In pharmacologically active doses, glucagon causes an increase in insulin secretion from healthy ?-cells of the pancreas.
Glucagon has an ino- and chronotropic effect on the heart due to an increase in the formation of cAMP (i.e., it causes an effect similar to ?-adrenergic receptor agonists, but without their involvement).
In high doses, glucagon induces strong intestinal relaxation not mediated by adenylate cyclase.
Pharmacokinetics
It is metabolized in the liver, kidneys, plasma, tissues. Metabolic products are excreted in the urine. T1 / 2 is 3-10 minutes.
Side effect
From the digestive system: nausea, vomiting.
Allergic reactions: skin rash, itching.
From the side of the cardiovascular system: rarely - tachycardia, transient increase in blood pressure.

It should be borne in mind that glucagon exerts its effect only in the presence of glycogen in the liver, therefore it is ineffective in patients on a reduction diet, with adrenal insufficiency, and with chronic hypoglycemia.
In patients taking beta-blockers, severe tachycardia and arterial hypertension are possible when glucagon is administered.
Drug interactions
Glucagon enhances the effect of oral anticoagulants. Phenytoin inhibits glucagon-mediated insulin release.
