DESCRIPTION

Apoquel 3.6 mg - oral tablets. In appearance, Apoquel is an oval white or almost white film-coated tablet with a dividing groove in the middle and an inscription on both sides of the tablet: AQ and S for a dosage of 3.6 mg.

COMPOSITION

As an active ingredient, oclacitinib maleate (in terms of oclacitinib) 3.6 mg, respectively, as well as auxiliary substances: microcrystalline cellulose, lactose monohydrate, sodium carboxymethyl starch, magnesium stearate, Opadry II White film former and purified water.

PHARMACOLOGICAL PROPERTIES

Pharmacotherapeutic group: a drug of the group of selective Janus kinase inhibitors (JAK).

Oclacitinib maleate, which is a part of the drug, in a therapeutic dose inhibits the functions of pro-inflammatory, pro-allergic and pruritogenic (pruritogenic) cytokines, dependent on the enzymatic activity of Janus kinase JAKI or JAK3, the targeted action on which allows to inhibit the key mechanisms of itching associated with allergy, and helps to eliminate the symptoms of local inflammation; the cytokines involved in hematopoiesis and dependent on JAK2 are not significantly affected.

With long-term use at higher doses, oclacitinib can cause suppression of lymphoid tissue, inhibit T-cell activation by suppressing the signal at the level of IL-2 interleukin receptors, which characterizes the pharmacological action of compounds of this class.

After oral administration of the drug, oclacitinib maleate is rapidly absorbed in the gastrointestinal tract (bioavailability is 89%), enters the systemic circulation, reaching a maximum plasma concentration in less than 1 hour, penetrates into most organs and tissues, is practically not metabolized, excreted from the body in unchanged form, mainly with urine, the half-life is 3-4 hours.

According to the degree of impact on the body, Apoquel is classified as a moderately hazardous substance (hazard class 3 according to GOST 12.1.007) with weakly expressed cumulative properties, in the recommended dose it does not have mutagenic, carcinogenic, embryotoxic and teratogenic activity.

PURPOSE

Apoquel is prescribed for dogs with dermatitis associated with allergies (contact, food, parasitic) in order to eliminate itching and reduce focal skin changes, as well as to prevent recurrence of atopic dermatitis.

DOSAGE AND APPLICATION

Apoquel is administered to dogs orally individually from the hand, or mixed with food, or injected into the mouth forcibly in a single dose of 0.4-0.6 mg of oclacitinib per 1 kg of animal weight twice a day, no more than 14 days; as maintenance therapy for atopic dermatitis - once a day at the same dose, but not more than 14 weeks.

The scheme and duration of the course of the drug use depend on the etiology and pathogenesis of dermatitis, the clinical state of the animal and are determined by the attending veterinarian.

Make sure the dog has completely swallowed the required dose.

Food intake does not affect the bioavailability of oclacitinib.

SPECIAL INSTRUCTIONS

When working with the drug, one should observe the general rules of personal hygiene and safety measures provided for when working with medicinal products for veterinary purposes. When working with the drug, it is prohibited to drink, smoke or eat. After finishing work with the preparation, wash your hands with soap and tap water.

People with hypersensitivity to the components of the drug should avoid direct contact with Apoquel. In case of allergic reactions or in case of accidental ingestion of the drug into the human body, you should immediately contact a medical institution (with you have instructions for using the drug and a label).

In case of an overdose of the drug, the animal may experience the following symptoms: depression, vomiting, diarrhea, local alopecia, thickening of the skin fold, the formation of scabs and crusts, interdigital cysts, swelling of the extremities. There are no specific antidotes, the animal is treated with symptomatic treatment.

With the combined use of Apoquel with antimicrobial, anti-inflammatory and antiparasitic drugs, no pharmacological interaction has been identified. Apoquel should not be used in conjunction with immunosuppressants and anticonvulsants.

It is not recommended to vaccinate during the treatment period using Apoquel, as well as 2 weeks before and after the treatment period.

The peculiarities of the action during the first use of the drug and when it is canceled have not been identified.

CONTRAINDICATIONS

A contraindication to the use of Apoquel is the individual intolerance to animals of its components. Do not use the drug in dogs with signs of immunosuppression, such as hyperadrenocorticism, as well as in dogs with progressive malignant neoplasms. You should not use the drug in males during the mating period, dogs under 12 months of age and / or weighing less than 3 kg, as well as puppies and lactating bitches.

It is forbidden to use Apoquel to bitches during pregnancy and lactation, as well as to puppies under 12 months of age.

SIDE EFFECTS

Side effects and complications with short-term use of Apoquel can manifest themselves as a depression, a disorder of the gastrointestinal tract function, cystitis; in animals receiving Apoquel for more than 16 days, approximately in 1% of cases, there may be an increase in appetite and aggression, skin and subcutaneous neoplasms, skin infections caused by yeast fungi, enlarged lymph nodes, exacerbation of demodicosis and oncological diseases.

SHELF LIFE AND STORAGE

The shelf life of the medicinal product, subject to storage conditions, is 2 years from the date of manufacture. The unused halves are stored for no more than 3 days by placing in a bottle. It is forbidden to use Apoquel after the expiration date.

Store the drug in the manufacturer's closed packaging, in a place protected from direct sunlight, separately from food and feed, at a temperature of 2 В° C to 25 В° C.

PACKAGING

Apoquel is produced in packs of 100 tablets in white dense polyethylene bottles with a screw cap made of polypropylene that cannot be opened by children. Each package of the medicinal product is accompanied by instructions for use in Russian.
