Pharmacological action

Nemozole - an anthelmintic drug. The main mechanism of action is associated with the inhibitory effect on the polymerization of beta-tubulin, which leads to the destruction of the cytoplasmic microtubules of the cells of the intestinal tract of helminths, changes the course of biochemical processes (inhibits glucose utilization), blocks the movement of secretory granules and other organelles in the muscle cells of roundworms, causing their death. Most active against larval forms of cestodes - Echinococcus granulosus and Taenia solium, nematodes - Strongyloides stercolatis.

Pharmacokinetics

Absorption and distribution of

After oral administration, it is poorly absorbed from the gastrointestinal tract, unchanged, it is not detected in blood plasma. Oral bioavailability is low, about 30%. Admission simultaneously with fatty foods increases the absorption and the value of Cmax in blood plasma by 5 times.

Metabolism and excretion

Albendazole is rapidly biotransformed in the liver into a primary metabolite - albendazole sulfoxide, which also has anthelmintic activity. Cmax of albendazole sulfoxide in blood plasma is achieved in the range from 2 to 5 hours, binding to plasma proteins is 70%. Albendazole sulfoxide is almost completely distributed throughout the body and is found in urine, bile, liver, in the wall of the cyst and cystic fluid, and cerebrospinal fluid.

Albendazole sulfoxide in the liver is converted to albendazole sulfone (a secondary metabolite) and other oxidized products. T1 / 2 albendazole sulfoxide - 8-12 hours. It is excreted in the form of various metabolites in the urine.

Pharmacokinetics in special clinical cases

Contraindications

retinal damage

Composition

1 tablet contains:

Active ingredient: albendazole 400 mg

Excipients: corn starch, sodium lauryl sulfate, povidone-30, methyl paraben, propyl paraben, citric acid, gelatin, purified gum talcum, sodium talcum colloidal silicon dioxide, magnesium stearate, aspartame, pineapple flavor, mint flavor.

Dosage and administration

Take with food. With neurocysticercosis and echinococcosis, patients weighing more than 60 kg should be taken 400 mg twice a day. For those patients whose weight is less than 60 kg - twice a day at 15 mg / kg - this is the maximum allowable dose of medication per day.

The duration of the course of therapy with Nemozol tablets can be from 1 week to a month. With echinococcosis, it is necessary to drink three courses of 4 weeks, while doing a two-week break.

Side effects of the digestive system

: impaired liver function with a change in liver function tests (mild or moderate increase in transaminase activity), abdominal pain, nausea, vomiting.

Drug Interaction

Concomitant administration of albendazole with irasiquantel, dexamethasone and cimetidine may increase the concentration of albendazole sulfoxide in the blood. Concomitant use with carbamazepine, phenytoin, phenobarbital and ginseng can lead to a decrease in the concentration of the drug albendazole in the gut.

Overdose

Treatment: gastric lavage, activated charcoal. If necessary, conduct symptomatic therapy.
